Pyrosequencing reactions take place on beads in tiny wells
(a) In the 454 sequencing system, single strands of DNA are replicated on tiny beads in preparation for sequencing. (b) The sequencing reactions of pyrosequencing take place in tiny wells arranged on plates. The many wells in a plate, and the very small reaction volumes, allow massively parallel sequencing of DNA at modest cost.
